Molecular basis of arrhythmias.

چکیده
The characterization of single gene disorders has provided important insights into the molecular pathogenesis of cardiac arrhythmias. Primary electrical diseases including long-QT syndrome, short-QT syndrome, Brugada syndrome, and catecholaminergic polymorphic ventricular tachycardia have been associated with mutations in a variety of ion channel subunit genes that promote arrhythmogenesis. Pathological remodeling of ionic currents and network properties of the heart critical for normal electrical propagation plays a critical role in the initiation and maintenance of acquired arrhythmias. This review focuses on the molecular and cellular basis of electrical activity in the heart under normal and pathophysiological conditions to provide insights into the fundamental mechanisms of inherited and acquired cardiac arrhythmias. Improved understanding of the basic biology of cardiac arrhythmias holds the promise of identifying new molecular targets for the treatment of cardiac arrhythmias.

Genetic and molecular basis of cardiac arrhythmias: impact on clinical management parts I and II.
Genetic approaches have succeeded in defining the molecular basis of an increasing array of heart diseases, such as hypertrophic cardiomyopathy and the long-QT syndromes, associated with serious arrhythmias. Importantly, the way in which this new knowledge can be applied to managing patients and to the development of syndrome-specific antiarrhythmic strategies is evolving rapidly because of the...
متن کاملThe Study of Various Cardiac Arrhythmias in Patients Poisoned With Aluminum Phosphide (Rice Tablet)
Background: The poisoning is mostly due to suicide and is observed in younger age groups. The systemic manifestations of ALP poisoning are common but pernicious manifestations of them are cardiotoxicity, shock, and ARDS. The clinical hallmark of cardiotoxicity is arrhythmia. The aim of this study was to investigate cardiac arrhythmias in patients poisoned with ALP who were admitted to Razi Ho...
متن کاملCompartmentalized regulations of ion channels in the heart.
The rate and force of contraction of the heart are precisely controlled by compartmentalized regulation of cardiac ion channels which determine electrical activities. It is known that modulation of cardiac ion channels, which is caused by drug administration, sympathetic nervous system stimulation and gender difference, can increase risks of lethal arrhythmias in carriers of inherited disease m...
متن کاملAnimal models for atrial fibrillation: clinical insights and scientific opportunities.
Atrial fibrillation (AF) is the most common arrhythmia in clinical practice. A variety of animal models have been used to study the pathophysiology of AF, including molecular basis, ion-current determinants, anatomical features, and macroscopic mechanisms. In addition, animal models play a key role in the development of new therapeutic approaches, whether drug-based, molecular therapeutics, or ...
